          【摘要】 目的 观察原位再生医疗技术(Moist Exposed Burn Therapy/ Moist Exposed Burn Ointment, MEBT/MEBO) 对大鼠慢性难愈合创面磷酸化ERK1 及c?myc 蛋白表达的影响?方法 将80 只SD 大鼠随机分为空白对照组(20 只)?MEBO 组(20 只)?贝复济组(20 只) 和模型组(20 只), 其中空白对照组大鼠仅作皮肤全层切除, 不予药物干预; MEBO 组?贝复济组及模型组在皮肤全层切除后, 建立慢性难愈合创面模型, 其中模型组建模后不予药物治疗, MEBO 组及贝复济组建模后分别采用MEBO 及贝复济换药治疗, 并于治疗第3?7?14 天,通过Western Blotting 技术检测4 组大鼠相同部位创面肉芽组织中磷酸化ERK1 及c?myc 蛋白的表达水平?结果 创面愈合过程中, 4 组大鼠创面肉芽组织中磷酸化ERK1 及c?myc 蛋白表达水平均呈先上升后下降的趋势?其中, 治疗第3?7?14 天, 空白对照组创面肉芽组织中p?ERK1 及p?c?myc 的表达水平均明显高于其他3 组, P均<0.001,差异具有统计学意义; 治疗第7?14 天, MEBO 组与贝复济组创面肉芽组织中p?ERK1 及p?c?myc 的表达水平均高于模型组, P均<0.05, 差异具有统计学意义, 但MEBO 组和贝复济组组间对比, P>0.05, 差异无统计学意义?

          【Abstract】 Objective To observe the effect of Moist Exposed Burn Therapy/ Moist Exposed Burn Ointment(MEBT/ MEBO) on the protein expression of phosphorylated ERK1 and c?myc in the chronic refractory wounds of rats.Methods A total of 80 SD rats were divided into four groups randomly: blank control group (20 rats), MEBO group (20 rats), rb?bFGF group (20 rats) and model group (20 rats). In the blank control group, the rats weren’t given any medication after the simple full?thickness skin excision, while in the other three groups, chronic non?healing wound model was established after the full?thickness skin excision. The wounds in the model group weren’t treated by any medication, while that in the MEBO group and rb?bFGF group were respectively treated by MEBO and rb?bFGF. On day 3, 7 and 14 of the treatment, the protein expression of phosphorylated ERK1 and c?myc in the wound granulation tissues from the same part of rats in each group were measured by the Western Blotting technique. Results During the wound healing process, the protein expression of phosphorylated ERK1 and c?myc both increased firstly followed by a decrease. On day 3, 7 and 14 of the treatment, the expression levels of p?ERK1 and p?c?myc in the granulation tissues of the blank control group were significantly higher than that in the other three groups, with P <0.001, showing a statistically significant difference. On day 7 and 14 of the treatment, the expression levels of p?ERK1 and p?c?myc in the granulation tissues of the MEBO group and rb?bFGF group were both higher than that in the model group, with P <0.05, showing a statistically significant difference.However, the comparison between the MEBO group and rb?bFGF group in this respect showed no statistically significant difference, P >0.05. Conclusion MEBT/ MEBO may promote the repair and regeneration of chronic refractory wounds by enhancing the protein expression of phosphorylated ERK1 and c?myc.
