Objective: To investigate the healing mechanisms of refractory acute β-ray-injured skin wound by observing the dynamic changes of cells expressing integrin β1 during the natural healing process of acute β-ray-injured deep second degree skin wound in rats. Methods: 96 SD rats were randomly divided into an experimental group (n=48) and a control group (n=48). Build a rat model of acute deep second-degree skin wound caused by β-ray in the experimental group; set up a rat model of deep second-degree skin wound of thermal scald in the control group. On 1d, 3d, 7d, 14d, 21d, 28d, 35d post-injury, record the wound changes and take wound tissue sample for HE staining and immunohistochemical staining to observe the histological changes and changes of cells expressing integrin β1 during the process of wound healing. Results: Compared with the control group, the wound healing time of rats in the experimental group was longer and the difference was significant according to the statistical analysis (P<0.01). The detection of integrin β1 showed there was a statistically significant difference between the two groups in terms of IOD value of cells expressing integrin β1 on day 1, day 3, day 7, day 14, day 21 post-injury (P<0.05), but the IOD value on day 28 and day 35 showed no statistically significant difference between the two groups (P>0.05). Conclusion: The healing time of acute deep-second degree skin wound caused by β-ray was relatively longer and the proliferation and differentiation of cells expressing integrin β1 were delayed during the healing process which could be one of the reasons why the healing of β-ray-injured acute deep-second degree skin wounds was delayed.
