          目的 研究探讨烧伤创面愈合后不同时期瘢痕组织中成纤维细胞DNA甲基转移酶1(DNMT1)的表达及意义。方法 收集烧伤创面愈合后不同时期的瘢痕组织标本(早期、增生期、消退期、成熟期)和正常皮肤组织标本作为研究对象,采用免疫组织化学方法检测瘢痕组织及正常皮肤组织中成纤维细胞内DNMT1的表达,并比较各组之间的差异;采用RT-PCR技术检测瘢痕组织及正常皮肤组织中成纤维细胞内DNMT1 mRNA的表达,并比较各组之间的差异。结果 瘢痕组织形成早期,成纤维细胞内DNMT1的表达水平逐渐升高(免疫组化评分:8.57±1.83),并于增生期达到高峰(10.41±1.58),消退期(7.17±1.19)及成熟期(4.90±0.88)逐步降低,而正常皮肤组织中成纤维细胞的表达呈阴性或弱阳性(0.81±0.87),组间差异具有统计学意义(P=0.000, P<0.01);RT-PCR技术检测的DNMT1 mRNA的表达水平在各组中的差异与免疫组织化学方法的检测结果一致(P=0.000,P<0.01)。结论 成纤维细胞DNMT1对烧伤创面愈合后瘢痕组织的形成过程发挥着重要作用,DNA甲基化可能参与了烧伤创面愈合后瘢痕的发展与演变。

          Objective To study the expression of fibroblast DNA transmethylase 1 (DNMT1) in scar tissues at different stages after the burn wound healing and its significance. Methods Collect normal skin tissue samples and scar tissue samples at different stages (early stage, proliferative stage, fading stage and mature stage) after the burn wound healing, andtestthe expression levels of fibroblast DNMT1 and DNMT1 mRNA in both scar tissues and normal tissues by making use of the immunohistochemistry technique and the RT-PCR technique respectively, followed by comparing the expression differences between the two kinds of tissues. Results At the early stage of scar formation, the expression level of DNMT1 in fibroblasts elevated gradually (immunohistochemical score being 8.57±1.83) and it peaked at the proliferative stage (10.41±1.58) but decreased gradually at the fading stage (7.17±1.19) and mature stage (4.90±0.88). By contrast, the expression of DNMT1 in normal skin fibroblasts showed negative or weakly positive (0.81±0.87) which had significant difference in statistics as compared with that in the scar tissue fibroblasts (P=0.000, P<0.01). The expression level differences of fibroblast DNMT1 mRNA between the scar tissues and normal tissues tested by the RT-PCR technique were consistent with that tested by the immunohistochemistry technique (P=0.000,P<0.01). Conclusion Fibroblast DNMT1 plays an important role in the formation of scar tissues after burn wound healing and DNA Methylation may participate in the development and evolution of the burned scar.
